Of course, you can do that by R clicking on the folder you want to password protect and then click on properties, in the properties window you can enable the encryption feature. This is the most normally used way to password protect folders on computer, while it's useless if you login your computer with admin account and then walk away, in this situation any people can access to the protected folder.
And folder protected by 3rd party software won't meet this problem. If you are interested in 3rd party file protection software, I recommend you to try Folder Protector, it's excellent in password protecting folders on computer as well as on flash drive.
